This is the penultimate main story quest in the game. It unlocks after completing the "Royal Misfortune" quest, and to start it, you must sail to Havana.
Edward pursues his third of three primary targets: Laureano Torres.

Quest stages of Tainted Blood
1. Follow Anne Bonny
Follow Anne Bonny. There are several soldiers in the area, but if you follow right behind Anne, you should not be spotted by them. Once you reach the roof of one of the houses near the church - where Rhona is waiting - a cutscene will trigger.
2. Meet with Anne Bonny
While you were talking to Rhona, Anne climbed the church to scout the area. You must join her.
Cross the rooftops toward the square where the church stands. You should see a guard ahead of you, and three more on the roofs surrounding the square. Since you are already here, assassinate all of them by moving clockwise across the rooftops.
When they are all dead, climb to the top of the church to speak with Anne.
3. Assassinate Torres
Torres and his bodyguards will be moving around the market square. You can use the zipline to reach the rooftops on the right side, then jump onto a nearby tree and wait there for Torres to come close.
When you strike the lethal blow, it turns out you killed Torres' body double - and after a short cutscene, you will be attacked by his chief bodyguard, El Tiburón.
4. Kill El Tiburón
You will not be fighting El Tiburón alone, as a few additional soldiers will periodically run in and attack you. They pose an added challenge, but they are also a source of extra ammunition, healing items, or can be used as human shields from El Tiburón's shots.
At the start of the fight, El Tiburón is well armored and immune to sword attacks, so your first task is to change that. You can destroy his armor using your pistols. Aim at him, and part of his armor will start glowing red. Shoot the glowing section to destroy it. You must destroy three parts in sequence: the left shoulder guard, the right shoulder guard, and the chestpiece.
Once you destroy El Tiburon's armor, you can face him in sword combat. El Tiburon uses only red attacks, which you must dodge. If you move too far away from him, he may shoot at you. Pistols are still a good weapon against him. Without part of his armor, El Tiburon will be faster - plus additional soldiers will keep spawning - so this fight will be a bit chaotic. Once you finally defeat him, a cutscene will trigger.
5. Reach the Assassin's Bureau
The bureau is located in the south of the city. It is best to travel across the rooftops to avoid being spotted by any soldiers - as you must be anonymous to speak with your friends waiting for you there. This quest will end, and the final main quest of the game will be unlocked - Ever a Splinter.
