This mission will be available starting from the middle of the fourth chapter of the game. To begin it, go to Valentine where on the porch in front of the general shop you will meet two brothers and a woman. Acrisius and Proetus want to prove which one of them is braver.
To check this, they will ask you to shoot at the bottles they will put on their heads. To do that, you will go to the backyard of the store.
Two brothers seeking adventure in the West, Acrisius and Proetus, have bottles shot off their heads to prove their bravery.

Objectives of Oh, Brother
1. Shoot the bottles
First, they will place the large bottles on top of their heads. Shoot the bottles and of course try not to kill the brothers.
That's not enough for them and after a while they will place the smaller bottles on top of their heads.
In the next round they'll come up with an additional obstacle. This time they'll stand on one leg.
When you're done, both brothers will take Helen home and this stage of the mission will be over.
2. Part II
Some time later you will meet Helen and her two admirers at the back of the Valentine Saloon. The brothers have a new idea of how to impress Helen. They want to show her that they can take the punches.
In order to prove their mettle brothers Acrisius and Proetus take punches in front of a local woman, Helen.
4. Kick Acrisius / Kick Proetus
But that will not be enough for them. So kick each one of them in the nuts.
The brothers will finally give up and go with Helen to heal the wounds.
5. Part III
Some time later you will be able to meet both brothers and Helen again. This time you will find them near the waterfall.
The brothers have another idea about how to prove to Helen their courage. They want to cross the waterfall in barrels.
7. Mount your horse / Take Helen to search for the barrels downstream
Get on your horse and drive down the river to a place where both barrels have been thrown ashore.
Luckily, both brothers survived.
They don't argue with each other anymore, because they finally realized how stupid they had been. The brothers don't want to have anything to do with Helen anymore and they walk away, reconciled to each other.
After floating over a waterfall in barrels and injuring themselves, the two brothers realize how foolish their attempts to prove themselves have been.
